Can I pray in clothes from a thrift store?

Yes, in most cases. Clothes from a thrift store are pure by default, so your prayer in them is valid. Wash them first for hygiene, and check for real impurities, pigskin items and unsuitable prints.
Pure unless you know otherwise
The basic rule is that things are pure unless you know they are impure. When a man worried that he had passed wind in prayer, the Prophet ﷺ said he should not leave the prayer unless he hears a sound or smells something (Sahih al-Bukhari 137). Scholars apply the same principle to clothes: doubt does not cancel certainty. You do not need to know who owned a shirt or what their religion was. In one report, Ibn Abbas was asked about skins brought by Magians, and he answered that the Prophet ﷺ said tanning purifies them (Sahih Muslim 366). Items from non-Muslim sources are not impure just for that reason. See Can I pray with a stain on my clothes?
Why you should still wash them
Washing is mostly about hygiene, not ruling. Second-hand clothes can carry dust, smoke, perfume and allergens, and a wash gets rid of all of them. A normal machine wash with detergent is enough for ordinary dirt. If you see or smell a real impurity, such as urine, vomit or blood, remove it before you pray in the item. Things worth checking
- Leather: tanned leather is pure, but avoid pigskin. Check labels on gloves, belts and linings.
- Prints: skip alcohol brands, crosses, occult symbols and offensive slogans.
- Men's silk and gold: vintage ties, shirts and jewelry may be real silk or gold, which are not for men.
- Fit and coverage: make sure the item covers what it should when you bow and sit.
- Underwear and swimwear: many people prefer to buy these new, which is a hygiene preference, not a rule.

Thrift shopping fits Islamic values
Buying second-hand also avoids waste, which the Quran warns against:
It saves money for rent, family and charity, and keeps clothes out of landfill. You can also donate your own unused clothes to people who need them. See Can I donate my old clothes as charity?
