Can Muslims play American football with a pigskin ball?

Yes, Muslims can play American football. Despite the nickname, official NFL and college footballs are made of cowhide leather, and most practice and youth balls are rubber or synthetic. Even if a ball were real pig leather, touching it wouldn't break your wudu.
"Pigskin" is just a nickname
The name comes from the early days of the game, when balls were often made from inflated animal bladders, sometimes from pigs. Today the official NFL ball is cowhide, and college and high school game balls are usually cowhide or composite leather. Cheaper balls for practice and the backyard are rubber or synthetic. So in almost every case, the "pigskin" in your hands has nothing to do with pigs.
What if the leather really is pig?
The Quran describes pork as impure:
The Prophet ﷺ said that when a hide is tanned, it becomes pure (Sahih Muslim 366). The Hanafi, Shafi'i and Hanbali schools, and the main Maliki view, exclude pig from this, so they consider pig leather impure even after tanning. A minority, including Abu Yusuf and the Zahiri scholars, say tanning purifies every hide. See Can Muslims wear pigskin leather?
Touching it doesn't break wudu
Touching something impure isn't one of the things that break wudu. Most scholars also say that dry contact with a dry impure object doesn't transfer impurity. If your hands are wet or sweaty and you know the ball is pig leather, simply rinse your hands before you pray. This works the same way as touching a dog. See Does touching a dog break my wudu?
If something impure gets on your kit, wash that spot before praying in it. See Can I pray with a stain on my clothes?
The sport itself
American football is a permissible team sport, just like soccer. The usual guidelines apply: men cover navel to knee (game pants do this well), keep tackles within the rules, avoid deliberate harm and trash talk, don't bet on games, and plan practice and game days around prayer.
